Use of dehydrosulphurenic acid for inhibiting the growth of cancer cells

Introduction
Antrodia cinnamomea (AC) is a fungus parasitic on aromatic tree Cinnamomum kanehirai Hay. (Lauraceae). It could only be found in Taiwan and has been widely used for many years as a functional food to protect against hepatoma and hepatitis virus infection in general population. It has been reported that extracts and several ingredients isolated from AC possess various pharmacological and biological activities. Fermented AC extract and adenosine isolated from AC prevented serum deprivation-induced PC12 cell apoptosis. Ethylacetate extract from AC fruiting bodies attenuated the cancer invasion of human liver cancer PLC/PRF/5PLC/PRF/5 cells and induced apoptosis of hepatoma Hep3B cells. Polysaccharides isolated from the mycelium of AC protected liver against injury in mice and suppressed angiogenesis. Though AC extracts were reported to have pharmaceutical effects from many studies, the application in growth inhibition of leukemia and pancreatic cancer cell was never reported. In order to identify the active ingredients for inhibiting cancer cell growth from the extracts of AC, the compound of the MMH01 was isolated and purified in this invention. The chemical name of MMH01 is 24-methylenelanosta-7,9(11)-diene-3β,15α-diol-21-oic acid, also called dehydrosulphurenic acid. Dehydrosulphurenic acid inhibits the growth of pancreatic cancer cell line BxPC-3 through induction of apoptosis. Dehydrosulphurenic acid also induces apoptosis and mitotic catastrophe to inhibit the growth of leukemia cell line U937. The present invention of MMH01 compound to inhibit the growth of tumor cells, which was further used as a medicinal or pharmaceutical composition to treat cancer cell or to enhance the therapeutic effects.

The present invention relates to a method for inhibiting tumor growth, in particular to the method using dehydrosulphurenic acid to inhibit the growth of leukemia cell or pancreatic cancer cell by a compound extracted and purified from AC. Dehydrosulphurenic acid of the invention can be used as pharmaceutical composition to inhibit the tumor growth of leukemia or pancreatic cancer.
