OBI Pharma unveils new ThiOBI and HYPrOBI technology platformsApr 30, 2025

OBI Pharma announced the debut of its innovative ThiOBI platform and HYPrOBI linker technology at the American Association for Cancer Research (AACR) annual meeting. These self-developed key technologies significantly enhance the development capabilities of next-generation antibody-drug conjugates (ADCs), leaving attendees impressed and eager to follow up with further inquiries.

OBI Pharma CEO, Hui-Chun Wang, stated that both the ThiOBI and GlycOBI platforms have their unique strengths, providing OBI Pharma with a comprehensive technological platform. Depending on the needs of their partners, the company will offer a variety of ADC development options, boosting OBI Pharma’s competitiveness in the global ADC market and meeting the diverse demands of the market.

The AACR annual meeting, held from the 25th to the 30th in Chicago, USA, brought together more than 20,000 cancer-related professionals from around the world. OBI Pharma presented seven posters during the conference, including the first public unveiling of the ThiOBI platform and the HYPrOBI linker technology.

Teng-Yi Huang, Senior Director of OBI Pharma’s Medicinal Chemistry Department, explained that the ThiOBI platform utilizes a cysteine-based (Cys-based) linkage method, which is currently the mainstream technology in ADC development. It has been clinically proven to have potential binding sites. The ThiOBI platform, combined with the irreversible cysteine conjugation technology and the HYPrOBI linker, offers a more stable and reliable technological solution for ADC development.

Additionally, OBI Pharma’s self-developed HYPrOBI linker technology features masking and shielding effects, addressing several challenges traditionally faced in ADC development. The masking effect reduces the hydrophobicity of the payload, enhancing its solubility, while the shielding effect creates a protective layer around the payload to prevent premature drug release in the bloodstream, ensuring effective release once the drug reaches the tumor site.
