In an exciting development, a team led by Vice President Tsung-Jung Ho at Hualien Tzu Chi Hospital has successfully harnessed modern biotechnology to extract active peptides from the traditional Chinese medicine formulation "Tortoise Shell and Deer Horn Two Immortals Glue," demonstrating significant potential in promoting bone health and regeneration.

Modern technology unlocks the secrets of Guilu Essence, active peptides show anti-Osteoporosis potential

Tortoise Shell and Deer Horn Two Immortals Glue has long been used clinically to treat various bone-related diseases. The team at Hualien Tzu Chi Hospital utilized LC-MS/MS (liquid chromatography-tandem mass spectrometry) to process commercially available Tortoise Shell and Deer Horn Two Immortals Glue products with pepsin, simulating human stomach digestion. They then used HPLC (high-performance liquid chromatography) to isolate the pentapeptide TSKYR, which stimulates bone cell proliferation.

Considering that TSKYR might further degrade into shorter peptide fragments such as TSKY, TSK, and YR in the small intestine, the team chemically synthesized these four peptides. They conducted cell proliferation tests, activity assays, and polymerase chain reaction (PCR) analysis to compare and confirm their effects on bone cells.

Peptides promote bone cell growth, chondrocyte proliferation increases by 47.5%

Experimental results revealed that active peptides from Tortoise Shell and Deer Horn Two Immortals Glue, including TSKYR, could stimulate the proliferation, differentiation, and activation of human osteoblasts and chondrocytes. Specifically, the tripeptide TSK increased the area and density of mineralized nodules by 30 and 50 times, respectively, and stimulated the growth of C20A4 chondrocytes by 47.5%. Additionally, the dipeptide YR showed a significant synergistic effect with calcium ions, promoting osteoblast proliferation, which is believed to aid calcium absorption in the body. These findings suggest that the active peptides in Tortoise Shell and Deer Horn Two Immortals Glue have the potential to prevent or treat Osteoporosis and Osteoarthritis.

A major breakthrough in Osteoporosis treatment

Professor Hao-Ping Chen from the research team noted that current Osteoporosis treatments often have side effects and are limited to diagnosed patients, while commercially available bone health supplements offer limited efficacy. The development of active peptides from Tortoise Shell and Deer Horn Two Immortals Glue marks a significant breakthrough in the treatment of Osteoporosis in Taiwan.

The research team has filed patents in Taiwan and China and is applying for a PCT to enter the European, American, and Japanese markets. They have commenced animal trials and are actively preparing for human clinical trials, aiming to develop these peptides into potential therapeutic drugs or preventive health supplements for Osteoporosis and Arthritis.
